CRISPR/Cas9-engineered inducible gametocyte producer lines as a valuable tool for Plasmodium falciparum malaria transmission research

Abstract The malaria parasite Plasmodium falciparum replicates inside erythrocytes in the blood of infected humans. During each replication cycle, a small proportion parasites commits to sexual development and differentiates into gametocytes, which are essential for transmission via mosquito vector. Detailed molecular investigation gametocyte biology has been hampered by difficulties generating large numbers these highly specialised cells. Here, we engineer P. NF54 inducible producer (iGP) lines routine mass production synchronous gametocytes conditional overexpression commitment factor GDV1. NF54/iGP consistently achieve rates 75% produce viable that transmissible mosquitoes. We also demonstrate further genetic engineering is valuable tool targeted exploration biology. In summary, believe iGP approach developed here will greatly expedite basic applied stage research.
